摘 要:
以拟南芥糖代谢相关酶基因序列作为探针, 对葡萄EST数据库进行同源检索筛选, 获得相应的同源EST序列, 并以葡萄果实cDNA为模板, 通过PCR反应, 对葡萄糖代谢途径相关基因进行了分析。进一步预测了葡萄糖代谢途径相关酶编码基因个数, 分别位于1、4、5、6、7、8、11、12、13、16、17、18、19号染色体, 另有3条序列未能定位到染色体, 并对编码基因表达强弱进行了分析, 为深入了解葡萄果实内糖代谢的机理提供一定的工作基础, 也可以为如何调控果实糖代谢以及提高葡萄果实品质提供理论依据。关键词:葡萄; EST; 糖代谢

Abstract:
Homologous grapevine EST sequences were searched against the grape EST database using corresponding sugar metabolism gene sequences from Arabidopsis as the probes. The related genes involved in the sugar metabolic pathway in grape was then analyzed by PCR reactions using cDNA prepared from developing grapevine fruits as PCR template. From the EST sequences obtained and verified, it is possible to predict the coded genes related to sugar metabolism and analyze gene numbers involved and the location of these genes locations on chromosomes 1, 4, 5, 6, 7, 8, 11, 12, 13, 16, 17, 18 and 19. Three sequences could not be linked on any chromosome. This study also analyzed the expression patterns of coded genes where it was found that expression varied with the stage of fruit development. The results of this study could provide a theoretical basis for further researches on regulation of fruit sugar metabolism and improvement of grapevine fruit quality.Key words: grape; EST; sugar metabolism
